Der gerade aus dem Knast entlassene Jake (John Belushi) und sein Bruder Elwood Blues (Dan Aykroyd) sind stadtbekannte Chicagoer Ganoven, die Schwester Mary Stigmata (Kathleen Freeman) Besserung gelobt haben. Ihr fehlen 5000 Dollar, um das Waisenhaus weiter zu betreiben, in denen die beiden aufgewachsen sind. Bei Reverend Cleophus James (James Brown) kommt Jake die rettende Idee: Sie versuchen ihre alte Band zu reaktivieren und ein Konzert zu geben.
